DNA replication is a highly regulated process in eukaryotic cells, and it is coordinated by the replisome, a multi-protein complex that ensures the correct duplication of the genetic material. Eukaryotic cells are also equipped with surveillance mechanisms coupled to the replisome to prevent the transfer of damaged DNA to the daughter cells. Our latest results provide novel insights into the critical mechanisms which control DNA synthesis and the DNA replication checkpoint by showing that CK2a is essential for maintaining the stability of the replisome machinery and for optimal activation of the ATR-CHK1-mediated signalling pathway upon induction of DNA replication stress.

The Lundbeck Foundatioon supported the postdoc project entiteled ‘Targeting splicing for the chemo-prevention of brain metastases’ The project aims to develop a therapeutic strategy against brain metastasis by altering splicing of thymidylate synthase (TS) mRNA using splice switching oligonucleotides (SSOs).

We applied metabolomics and lipidomics to examine temporal regulation of metabolism during starvation in wild‐type Caenorhabditis elegans and in animals lacking the transcription factor HLH‐30. Our findings show for the first time that starvation alters the abundance of hundreds of metabolites and lipid species in a temporal‐ and HLH‐30‐dependent manner.

Cellular plasticity-regulating process of EMT as a model to show the efficacy of a fluorescent sensor to separate cells with distinct EMT signatures, based on miR-200b/c activity. The system was combined with a CRISPR-Cas9 screening platform to identify miR-200b/c upstream regulating genes. The sensor allows to infer miRNAs properties, as profiling of sorted cells indicated miR-200b/c as a molecular switch between EMT differentiation and proliferation, and suggested a role for metabolic enzymes in miR-200/EMT regulation.

ACBP-/- mice and skin-specific ACBP-/- mice exhibit increased energy expenditure, increased food intake, browning of the iWAT, and resistance to diet-induced obesity. The metabolic phenotype, including browning of the iWAT, was reversed by housing the mice at thermoneutrality or β-adrenergic blocking. Interestingly, these findings were phenocopied in flaky tail mice.

In this study, we show that the splicing factor SRSF10, which is particularly abundant in neuronal tissue, binds to ISS-N1, and that the long and short isoforms of SRSF10 inhibit SMN splicing with different strength correlating with the length of their RS domain. Our results suggest that the ratio between the SRSF10 isoforms is important for SMN splicing regulation.

Best Christmas Decoration 2020
This years contest of decorating the best Christmas office at the Department was expanded to also include home offices. Thomas from the Translational Biology section did a great job decorating his home office. Therefore he won the honorablel 'Gertrud Sand' price 2020.

Today we met for a corona friendly social activity in the Translational Biology section. We played bingo online on zoom with Thomas as a great bingo host, many nice prices and great time together, while separated.
