LONG LIFE Family Study

Danish Aging Research Center

The LONG LIFE Family Study (LLFS) is a collaborative research study, funded by the National Institute on Aging (NIA) and includes prestigious institutions from the U.S. in St. Louis, New York, Pittsburgh, Boston, and the SDU in Denmark. LLFS is studying families from the U.S. and DK with multiple members who have reached a very old age. The LLFS Project began in 2005, when the collaborating institutions began formulating the Study Design. In 2006, participant recruitment was launched and continued through June of 2009.

In Phase 1 of the study a total of 4,923 visits were completed by the LLFS Field Centers. The total number of families enrolled in the study are 535, consisting of 1,499 individuals in the Proband Generation, 2,594 individuals in the Offspring Generation and 830 Spouse Controls. In DK 1,268 individuals from 76 families participated.

Phase 1 survey dataset consisted among others of information about: Family, Health, Medication Inventory, Hospitalizations, Self-rated Health, Activities of Daily Living (ADL), Education, Occupation, Living Habits, Personality (NEO-FFI) and Depression (CES-D). Physical ability was tested by a short physical performance battery (SPPB): Balance Test, Gait Speed Test, Chair Stand Test and Grip Strength Test. Blood Pressure, Spirometry, and Anthropometry were measured as well. Finally, a cognitive battery consisting of Folstein Mini-Mental State Examination, Logical Memory, Digit Span, Fluency and Digit-Symbol Substitution Task was assessed. In addition, blood was drawn for analyses of various biomarkers, such as: Creatinine, Glucose, Cholesterol (LDL, HDL), CBC, Platelets, Lymphocytes and DNA.

In Phase 2, genotypic and phenotypic analyses of the large amount of data were conducted, and all surviving participants completed an annual follow-up interview in the years 1, 2, 3, 4, 5, and 6 after the initial in-home assessment. The interview focused on ADL-functions, psychological and physical health, health habits and sleep.
